My primary research interests are focused on novel loop design in TIM barrels. TIM barrels are of particular import due to their overrepresentation in enzymes; to date, at least fifteen distinct enzyme families use TIM barrels to establish catalysis, making TIM the most common enzyme fold known. The general ubiquity of TIM barrels make them an attractive target for protein design. Current projects are focused on generating a TIM barrel with non-native binding affinity via sampling and scoring grafted CDR-H3 loops using Rosetta. The resulting candidate structures will then be evaluated for experimental validity.
