My first project will give rise to a new technology platform to study membrane protein stability in a lipid-based membrane mimetic. Membrane protein analysis is complicated by membrane protein hydrophobicity, making stability testing challenging to study. I am utilizing nano Differential Scanning Fluorimetry (nanoDSF) and an engineered non-fluorescent ‘ultra-dark’ nanodisc (replaces both tryptophan and tyrosine resides with non-fluorescent sidechains) to assess membrane protein stability.
My second project is to experimentally determine the membrane orientation of SMIM10L1, a mitochondrial membrane protein that is of therapeutic interest because it downregulates melanoma cell proliferation. Prior computational research through Rosetta has already shown an alpha helical character and identified two potential orientations, and I am experimentally determining the orientation utilizing NMR studies.
